Hydroxyproline (Hyp) is the major component of the protein collagen comprising 13.5% of mammalian collagen. It is formed post-translationally by the hydroxylation of specific Proline residues by prolylhydroxylase enzyme. Dysregulation in collagen metabolism may result in pathologies such as fibrosis (too much collagen) or osteoarthritis (too little collagen). Therefore measurement of collagen production is important in many disease related studies. Hydroxyproline in tissue hydrolysates can be used as a direct measure of the amount of collagen present.

BioIntersect’s assay provides a simple, easy-to-perform, quantitative method of measuring Hydroxyproline. This assay can reliability detects upto ~3 μM of Hydroxyproline or 0.1 μg per well.
